Comparative Studies on Brucella Antigen

Abstract: Due to the spreading of brucellosis in Egypt in the last few years, the need for a rapid diagnosis test antigens for screening tests increased. In the present work trials were done to produce these antigens locally and to compare it with the imported ones.
Rose Bengal Plate test antigen (RBPT) was prepared from S 1119/3 and S 99 according to the protocol of joint FAO/WHO.
Buffered acidified Plate (BAPA) was prepared from S 1119/3 and S 99 according to the protocol of ames Iowa.
Serum Agglutination Test antigen (SAT) was prepared from 1119/3 and S 99 according to the protocol of Ames Iowa , beside the SAT from S 99 the only antigen produced in Egypt according to the protocol of joint FAO/WHO.
Using all these antigens, 110 cattle serum samples and 15 sheep serum samples were examined and the results were as follows :
-It was found that there were a complete similarity between all the antigens locally prepared and imported including RBPT, BAPA and SAT.
- Using RBPT it was found that number of cattle positive were (69.09%) suspect were (10.00%) and negative were (20.91%).
-Using BAPA the results were (70.92%) positive, (10.90%) suspect and (18.18%) negative.
-In case of SAT antigen, the different way of interpretation CVL, FAO and USDA were discussed.
-The results of SAT were (66.37%) positive, (18.18%) suspect and (15.45 %) negative.

The differences between the different antigens in diagnosing brucellosis were discussed, also the sensitivity and accuracy of all tests were reviewed.
The harmonic coefficient (R) varied from 0.70 to 0.74. Similar results were obtained when these tests were applied on sheep serum samples.
From theses results, it is clear that there is a complete similarity between the locally prepared and imported antigens in terms of sensitivity, reproducibility and aaccuracy and hence were very valuable in the diagnosis of brucellosis.
In conclusion the author looks forward that the authorities will support the production of such antigens on a large scale to cover the need of the country.
