Production of attenuated sheep pox vaccine from the Egyptian strain

Abstract: The Egyptian strain of sheep pox virus was purified and adapted on tissue cultures, lamb testicle cells and Vero cell line, for 27 passages till reached the clearest cytopathic effect (CPE) after 5 days post inoculation with production of a highest titre of virus (10 6.1 TCID50/ml). For vaccine production the tissue culture adapted virus was added to an equal volume of lactalbumen sucrose stabilizer and lyophilized, the lyophilized vaccine had a titre equal to 10 5.5 TCID50/ml in tissue culture and 10 5 SID50/ml in susceptible lambs. The prepared vaccine passed successfully the sterility, safety and potency tests, as well as the duration of protection acquired from vaccination was also estimated and proved that the vaccinated lambs with the prepared Egyptian tissue culture sheep pox vaccine resisted challenge with the virulent sheep pox virus for 10 months post vaccination (period of study). The humoral antibody level in the serum of vaccinated lambs was measured by the virus neutralization test (VNT) and enzyme linked immunosorbent assay (ELISA) and proved that the antibodies against sheep pox virus could be detected from the 1st to 4th week post vaccination and persisted for up to 3 weeks post challenge.
