Microsatellite DNA fingerprinting in comparison with conventional ways for evaluation of stability of Brucella Abortus strain99

Abstract: Brucella abortus strain 99 (S-99) was used for the preparation of different Brucella antigens used for the serological tests. The stability of the phenotypic and genotypic characterization is the principle factor for preparation of high quality Brucella antigens from this strain.
The protein profile and polymerase chain reaction (PCR) assay eight base pair varialbe number tandem repeats besides the phenotypic propertites included colony morphology, growth rate, biochemical reaction and sensitivity to antibodies and dyes were developed throughout ten successive bacterial passages. Loe percentage of colonial roughness and non significant changes in biochemical reaction and antibiotic sensitivity were observed after five passages.
Concerning the protein profile and the DNA fingerprinting no change was observed for the protein analysis, while there was different DNA fingerprint pattern after the seven passage.
The variable number tandem repeats (VNTRs) PCR could be used as a complementary test to evaluate Brucella starins and could substitute the conventional methods in the future.
