Preparation of rift valley fever virus hyperimmune serum using differnt methods of virus purification

Abstract: In this study hyperimmune serum against Rift Valley Fever virus (RVFV) was prepared in mice using two methods of virus purification (ultracentrifuge for purification of the virus in the first method and blotting the virus fraction on the nitrocellulose membrane for the second method).

Agar gel precipitation (AGPT), serum neutralization (SNT), ELISA and Western Blot were used to evaluate the hyperimmune serum. The highest dilution was 1:10240 for SNT and ELISA and 1:5000 for western Blot.

The results indicated that the hyperimmune sera were of high sensitivity and specificity and could be used successfully for diagnosis of Rift Fever disease and for evaluation of produced vaccine
