Evaluation the antigenic properities of camel pox virus (private)

Abstract: The growth of camel pox virus (CPV) was studied in Vero cells by evaluation its cytopathogenic effect (CPE) and estimation of the virus titre. CPE appeared at 48 to 96 hour post inoculation (PI) and characterized by aggregation and cell rounding which lead to presence of intercellular space grape-like clusters of refractile cells ending by lysis of the cell sheet.The virus titre was investigated in cell free (CF) and cell associated (CA) virus as well as the plaque technique. The titre of the virus was 5 log10 5.2log10 TCID50/ml in CF and CA virus respectively and 5 x 105 plaque forming unit (PFU). The viral protein was estimated by spectrophotometer and the protein profile was studied by sodium dodecyl sulphate polyacrylamide gel (SDS-PAGE) and Western blot. The total viral protein was 0.58gm % and the molecular weight (MW) of such viral proteins were found to be ranged between 31 to 105 kDa while the viral immunogenic proteins were 31, 34 and 35 kDa . The viral plaques were picke for the preparation of specific, potent and pure antigen which used in AGPT and dot ELISA for diagnosis of camel pox.
