Preparation of immunoglobulins conjugation with fluorescein isothiocyanate specific for fowl pox virus

Abstract: A study was applied to prepare a specific fowl pox hyperimmune serum conjugated with fluorescein isothiocyanate (FITC) which could be used in fluorescent antibody technique (FAT). The hyperimmune serum was prepared in SPF susceptible chickens reared in isolators. The antibodies were titrated and screened from any contaminants of avian origin by agar gel precipitation (AGPT), enzyme linked immunosorbent assay (ELISA), haemagglutination inhibition test (HI).
The immunoglobulins were precipitated by ammonium sulphate and conjugated with FITC. The conjugate was tested against infected and non-infected chorioallantoic membrane with fowl pox virus by FAT, it produced positive greenish yellow fluorescent reaction with infected specimens, till the dilution (1:400) while it gave negative reaction with non – infected ones. The prepared conjugate was evaluated against field – infected samples from fowl, pigeon and turkey suspiciously infected with avian pox disease. The results proved the sensitively of the conjugate to react with any part of the avian pox viruses, as well as its specificity to fowl pox virus is more than pigeon and turkey pox viruses.
