Isolation and characterization of fowl pox virus from naturally infected fowl and comparative studies between the new isolate and beaudette vaccinal strain.

Abstract: Local new field isolate of FPV was isolated from naturally infected chicken, obtained from private poultry farm in Cairo. It was found that it produced pock lesions on the choroallantoic membrane of ECE. To confirm that the isolate belongs to FPV the field isolate, was inoculated in different species of poultry as chickens, ducks, turkeys and pigeons. There are specific lesions at site of inoculation by using wing web method in pigeons, on other hand the virus induce generalized pox lesions which appear in turkeys and chickens while ducks did not produce any reaction. Another confirmation by using AGPT, clear lines of precipitation appears between the FP antiserum and the FPV field new isolates. Structural polypeptides of Beaudette vaccine strain and field isolate of FPV were analyzed on SDS- PAGE in 12 % poly acrylamide gel 15 and 21 polypeptides, ranged from 297.87 and 10.04 KDa with a total MW of 1081.01and 1712.64 respectively for the two viruses. So, the results shows some variations of viral protein profile between the two strains.
