Studies on canine hepatitis in Egypt I-isolation of the local strain and its propagation in cell cultures

Abstract: A ten –years old dog was came to a clinic suffering from off- food depression, ocular opacity in one eye, diarrhea and its urine had a sheep orange colour. It received a sympotomatic treatment for 3 days then died. It was subjected to post mortem examination, histopathological studies and trial for isolation and identification of the causative agent using different serological tests. The post mortem findings and histopathological examinations showed the presence of characteristic signs and lesions of infectious canine hepatitis. On the other side, trials of virus isolation and identification using haegglutination (HAT), virus neutralization (VNT), agar gel precipitation (AGPT), and fluorescent antibody (FAT) tests revealed that the causative agent was infectious canine hepatitis virus (canine Adenovirus-1, CAV-1). The obtained isolate was labeled as CAV- 1-Abbassia / 2002. Experimental infection of susceptible dogs with the obtained isolate showed typical symptoms of canine hepatitis and died within 10 days post infection showing characteristic post mortem findings and positive virus recover. The virus was propagated in some cell lines including African green monkey (VERO), baby hamster kidney (BHK), Made in Derby Bovine kidney (MDBK) and chicken embryo rough (CER). The MDBK were found to be the most suitable cells yielding the highest virus titer followed by VERO then BHK and lastly CER cells
