Studies on rinderpest virus in theileria annulata infected lymphoblastoid cells

Abstract: A vaccine strain of rinderpest virus (Kabete O) was passaged for three successive times in Theileria annulata infected lymphoblastoid cells. The virus showed its characteristic cytopathic effects in T. annulata infected cells without any effect cells on the schizont. Specific cell aggregations, intracytoplasmic inclusions and cyncytia formation appeared within 48-72 hours post inoculation. Immunofluorescent specific rinderpest intracytoplasmic antigen within 24-48 hours post inoculation. Giemsa staining showed healthy schizonts before infection with the rinderpest virus vaccine and its prescence as intact schizontal bodies in the cells during the viral infection of their host cells. The virus titre reached 107 TCID50/ml by the third passage with reduction in the time of harvestation from 7 days (for the 1 st passage) to 3 days (for the 3rd passage). This work planned as a preliminary study to produce a combined tissue culture vaccine against rinderpest and T. annualata.
