Comparative studies on the growth behaviour of peste des petits ruminants (PPR) virus in different cell cultures

Abstract: Pest des petits ruminants (PPR) virus was propagated for five successive passages in VERO cells, lamb kidney (LK) and calf kidney (CK) cell cultures. The virus titre of the fifth passage in VERO LK and CK was 5.7, 3.8 and 3.3 log10 TCID50 /ml. respectively. The cytopathic effect (CPE) of the virus of the fifth passage started on the 2nd , 4th and 5th day post inoculation and completed on the 4th, 6th and 8th day post inoculation in VERO, LK and CK respectively. Hematoxyline and Eosin staining (H&E) of infected cells showed that the CPE was characteristic and similar in all cell cultures. CPE changes were in the form of cell rounding, increasing refractibility, grap appe-arance, cytoplasmic and nuclear degenerations, cytolysis with fine anastomosing cell processes and multinucleated giant cell formation. This work could be a preliminary study to choose the suitable cells for vaccine production.
