Application of PCR as a diagnostic tool of clinical mastitis in dairy cattle in comparison to traditional methods

Abstract: Commercial dairy farms had serious mastitis problems in spite of its control hygienic measures, for detection of this problem, a total of 3000 and 1500 lactating cows& buffaloes, respectively, were examined for clinical mastitis in 8 different farms at Ismailia Governorate. 130 milk samples were taken from 108 dairy animals suffering from clinical mastitis. The Bacteriological examination revealed that 56(43.1%), 34(26.2%), 8(6.1%), 4(3.1%) and 4 (3.1%) from clinically mastitic milk samples were positive for Staphylococcus epidermidis, staphylococcus aureus, Streptococcus uberis. Bacillus anthracoid and Enterobacter agglomerans, respectively. While 24(18.5%) of the samples were negative. Polymerase chain reaction (PCR) was also conducted to Staphylococcus aureus isolates as amajor pathogenic bacteria in cases of mastitis, Genomic DNAs extracted from all S. aureus isolates (n=34), were successfully identified by this PCR assay. PCR aws proved to be specific and sensitive in diagnosis of clinical mastitis.
