Molecular evaluation of some antigenic extracts of Corynebacterium pseudotuberculsis and utility in serodiagnosis

Abstract: Virulence factors of C.pseudotuberculosis, represented by concentrated exotoxin containing culutre filtrate and whole cell lysate (WCL) FROM 3 local isolates of ovine, caprine (serotype I)and buffalo (serotype II) origins, were comparatively analyzed for their antigenic structure and cross-reactive determinants, using sodium dodecyle sulphate-polyacrylamide gel electrophoresis ( SDS-PAGE) and western imunoblotting. Morever, their utility as candidate antigens for serodiagnosis of C.pseudotuberculosis infection in Egyptian sheep and buggaloes was investigated, by an indirect enzyme-linked immunosorbent assay (ELISA) and agar gel immuno-diffusion test (AGIDT). SDS-PAGE analysis of the exotoxins revealed high similarity among the 3 local isolates as determined at protein subunits with molecular masses(Mr) of about 68, 40-41, 39, 31.5, 29, 27 and 20 Kilodaltons (kDa). While, 4 antigens with approximate Mr of 68, 40, 3.15-31 and 29 kDa immunoreacted (ovine) and cross reacted (caprinsand buffalo) in immunoblotting. The exotoxin of ovine isolate showed othr 2 immunoreactive antigens at Mr. of about 20 and 14 kkda. Concerning the WCL, no markded differences could be observed among the 3 local isolates. as numerous immunoreactive/ cross-reactive antigens were estimated at aproximate Mr of 120, 100, 90, 72-68, 64, 40, 31, 29 and 20 kDa. Both WCL and exotoxin antigenic extracts from the local ovine isolate proved effective potential candidates in ELISA and AGIDT for serodiagnosis of clinical and inapparent infections.
