The production and evaluation of peroxidase antibody conjugate for immunoassays to pest des petits ruminants virus (PPRV) by using goats

Abstract: PPR hyper immune serum with neutralizing antibody titer of 1024 was prepared in goats. The immune globulins were precipitated from this serum by a saturated ammonium sulphate solution. The amount of these globulins was found to be 12.5gm/dl and adjusted to be 18mg/ml of phosphate buffer then conjugated with horse radish peroxidase using periodate method. The induced PPR neutralizing antibodies in vaccinated sheep were estimated using serum neutralization test (SNT) and the indirect immunosorbent assay (ELISA) on the 3rd week post vaccination. SNT showed that vaccinated sheep acquired specific PPR neutralizing antibodies of a mean titer of 36. It was found that the optimum dilution of the imported conjugate was 1:5000 while that of the prepared conjugate was 1:6000 as determined by using known +ve PPR antigen; -ve PPR antigen and indirect ELISA was carried out on the same sheep serum samples revealed mean absorbance values of 0.105 and 0.109 using the imported and prepared conjugate respectively indicating the validity of the last one as a local product of good quality.
