The production and quality estimation of a peroxidase conjugated antibodies for immunoassays to rinderpest virus

Abstract: A preliminary study was conducted to prepare peroxidase conjugated rinderpest antibodies to be used in ELISA. These antibodies were prepared in white Newzeland rabbits and conjugated with horse radish peroxidase using periodate method which known as the most popular, easier and cheapest one. The titer of the prepared antibodies was found to be 1024 and the immunoglobulin value was 12.5gm/dl where its concentration was adjusted to be 18mg/ml. Serum neutralization test carried out on 10 serum samples obtained from experimentally vaccinated cattle, revealed that these animals exhibited rinderpest neutralizing antibodies with a mean titer of 56 by the third week post vaccination. The prepared conjugate was evaluated through the application of the indirect ELISA on the same cattle sera in order to estimate rinderpest antibodies in these samples. It was found that the optimum dilution of the final preparation was found to be 1:8000 while that of the imported conjugate was 1:4000. The mean obtained absorbance values were 0.0501 and 0.060 using the prepared and imported conjugate respectively. The results of ELISA appear to be parallel to those of serum neutralization test carried out on the same serum samples
