Optimization of some factors for Improving PPR vaccine

Abstract: Three virus –culture interacting aspects were investigated aiming for producing a PPR virus vaccine with superior quality. It has been found that the use of virus inoculums containing multiplicity of infection (MOI) 5 per cell , the utlization of rolling culture system and the inoculation of the virus on cell suspension were the best conditions under which maximum PPR virus titer on vero cell could be achieved. These data were persistently confirmed through the production of three successive PPR pilot batches possessing at least 6.5 log10 TCID50 /ml on vero cells. It has been deduced that vero cells at logarithmic phase are more susceptible to the virus infection than the mature ones. Also , the use of (MOI) 5 per cell with the rolling system procure rapid and continuous cell infection with the versatile virus while the lower (MOI) per cell with the stationary culture method necessitates more time to get maximum virus titer during which the cells become older and more reluctant to the virus infection.
