Preliminary study for preparation of serological and diagnostic conjugated ‎fluorescent hyper immune serum against fowl spirochaetosis.‎

Abstract: A specific hyperimmune serum was prepared in chickens and rabbit and ‎precipitated with ammonium sulphate. The obtained immunoglobulins lgG ‎were conjugated with fluorescein isothiocyanate for performing of ‎fluorescent antibody technique ( FAT) for serological diagnosis of avian ‎spirochaetosis in - vitro. It was found that using of methyl alcohol ‎‎(methanol) in fixation of berrelia anserine was better than using acetone. The ‎conjugated hyperimmune serum prepared in chicken gave positive reaction ‎up to dilution 1:1024 in the direct technique in comparison with that ‎prepared in rabbits which gave positive reaction up to 1:512 dilution. Results ‎of the indirect technique were better than that obtained by using direct ‎technique due to absence of the non-specific reaction in the indirect ‎technique. Non –infected samples showed a black back ground in negative ‎reaction while positive reaction provides the specific yellow green (apple ‎green) light under fluorescent microscope. ‎
