Trial for Minimization the Antifertility Action and the Genotoxicity of Diazinon to Male Rats by Using Different Patterns of Dipping

Abstract: The present studies were carried out to investigate the reproductive, hormonal and enzymatic effects of diazinon on mal rates using dipping and spraying methods. Histopathological changes in liver, testis, kidney, muscles and brain were also studied. Moreover tow cytogenetic endpoints -chromosomal aberration and micronucleus test- were used for evaluation the genotoxic effect of diazinon. Data revealed that diazinon toxicity induced obvious reproductive alterations. At the same time, diazinon showed that evoked significant increase in the percentage of dead sperm and tail & head sperm abnormalities. Diazinon caused significant reduction effect of testosterome level of the mature male treated rats. As for the effect of diazinon on enzymes activity, revealed significant inhibition of serum cholinesterase and glutathion S-trnsferase and brain cholinesterase activity were appeared. The histopathological data showed that diazinon exhibited histopathological changes in liver, kidney and brain. this accompanied by histopathological changes in liver, testis and brain but renal and muscular tissues appeared normal. These results revealed ascending arrangement by increasing the frequency of chromosomal aberrant cell starting from G1 to G6. The data showed that exposure to diazinon -G6- elevated that frequency of total number of the micronucleated PCE cells when compared with the other five groups of treated rats. The statistical analysis also revealed that the induction of micronucli in bone marrow cells of rats treated with the different patterns of dipping was decreased gradually from G5 to G1. The adverse effects noticed with dipping rats in detergent only explain why the pattern of this dipping was not achieved protective role in diazinon intoxication. This resulting from its own toxicity. on the other hand, the protective role noticed with dipping rats in water after dipping in diazinon against the diazinon intoxication may be due to its dilution by water. This dilution lead to decreased its concentration on animal body
