Abstract: One hundred and eighteen chicken tracheal swabs as well as serum samples were collected from different flocks at Matrouh and Sidi-Brani ELISA positive reactions were 36.44 % for Mycoplasma gaillisepticum (MG) and 8.47 % for Mycoplasma synoviae (MS)
The suitability of.polyrnerase chain reaction (PCR) for the detection of either Mycoplasma gallisepticum (MG) or M. synoviae (MS) was compared with cultural method. The culture was positive for 8.47 % of the examined swabs, while 12.7! % of the samples were found to be positive by PCR.
Intraspecies genotypic heterogeneity among isolates and strains of Mycoplasma gallisepticum and M. synoviae was demonstrated with the arbitrarily primed polymerase chain reaction (AP-PCR) method. The differences according to the banding patterns of their amplified DNA on agarose gels were characteristic for specific isolates. The method can be used to determine The similarity between isolates of MG and MS from various sources
