Antigenic variability among mycoplasmasisolated from cattle, buffaloes and camels BY sds-page and immunoblot

Abstract: Thirty six reference and field strains were subjected to sodium dodecyl sulfate-polyoesylamide gel electrophoresis (SDS PAGE) and immunoblot. The results of SDS-PAGE showed that there was no clear difference in the protein profiles of field isolates from cattle and those from buffaloes. It was also found that the homologous reference strains and field isolates showed similar molecular weights from 36.7 to 35.3 (KDs) in M. bovis, 83 to 10 KDs in M bovigenitalium, 83 to 43 KDs mM bovirhinis, 127 to 32 KDs mM arginini. The glucose positive isolates from camels had exactly similar molecular weights (96.1-22.7 KDs). Camel isolates showed similar protein bands at 96.1, 86.6 and 53.7 KDs as those of M. bovirhinis.
Mycoplasma bovis and M bovigenitalium imtnunoblots using M bovis antisera revealed that theM bovis strains have five immunogenic bands (92.9, 63.5, 48.8, 43.5 and 30.9 Kds). WhileM bovigenitalium strains cross reacted withM bovis antisera and showed two common bands (63.5 and 43.5 KDs). On the other hand, Mycoplasma biogenitalium strains reacted with homologous antisera and showed nine immunogenic bands. However, M. bovis strains cross reacted with !vL biogenitalium antiseraand showed two common bands (63.5 and 43.5 KDs).
The SD S-PAGE and immunoblot can help in the identification of glucose positive mycoplasmas isolated from camels as M bovishinis. Both tests are veiy sensitive and specific for typing of other mycoplasmas isolated from cattle and buffaloes.
