Evaluation of locally prepared elisa kits fordetection and differentiation betweenmycoplasma galliseticum andmycoplasma synoviae specific antibodiesin chicken serum

Abstract: Indirect enzyme-linked immunosorbent assay (ELISA) was developed for detection of Mycoplasma gallisepticum (MG) and M. synoviae (MS) specific antibodies in chicken sera. Antigen concentrations of 12.5 and 625 ig / well for MG and MS respectively gave the best absorbance values of positive and negative sera. Serum dilution of 1: 100 showed the least non-specific bindings and the large difference between positive and negative sera 1:5000 and 1:10000 dilutions were chosen as the standard conjugate dilutions for MG and MS respectively. The use of phosphate - buffered saline containing 0.05 % Tween 20 for washing and diluting steps resulted in optimal differentiation between homologous and het erogolous sera. Serum chickens 8620 from commercial poultry flocks were tested for MG and MS; serum plate agglutination test was the highest (51.8% and 28% for MG and MS), the ELISA test was 50.2 % and 26.4 % while the HI test was the least (47.3 % and 25% for MG and MS). The results of the present study proved that ELISA was more accurate than HI for confirmation of Mycoplasma gallisepticum and Mycoplasma synoviae infection.
