Immunogenicnitiy of some protein fractions of mycobacteria
Abstract: Purification of 45/47kDa protein complex from culture filtrate of BCG by Si200 column, six major fraction collected, 45/47 kDa present in fraction 2 , protein content was 1 mg/ml. Whole culture filtrate of M.bovis prepared. By SDS-PAGE analysis, there were band of 45/47 kDa in culture filtrates (M.bovis, BCG) harvested afetr 14 days. Vaccination of guinea pigs by 45/47 kDa, BCG, cultures filtrates (M.bovis, BCG) challenged afetr 12 weeks. Vaccines were evaluated by humoral immune response (indirect ELISA), cell mediated immune response (lymphocyte proliferation assay expressed by MTT). Studies of protective immunity: survival rate, root specificlung weight (RSLW). Histological appearance postchallenge by 28 days. Protection against tubercles infection cannot be guaranteed by 45147kDa alone.
