Comparative studies on attenuated and inactivated Rift Valley Vaccines

Abstract: RVF is acute infectious viral disease affecting many species of animals and man. Animal vaccination is the most important control measurement in endemic areas. The inactivated and attenuated RVF vaccines were used. The inactivated and attenuated RVF vaccines were used. It is important to compare between both types. Firstly the two vaccines were inoculated in two groups of sheep including pregnant ewes. Neither thermal reaction nor abortion were recorded all over period of experiment, except that there was an elevation of body temperature for one day post vaccination with attenuated vaccine. T-cell response as cellular immune response of sheep vaccinated with live attenuated vaccine was significantly high at 1-5 week post vaccination with attenuated vaccine. T-cell response as cellular immune response of sheep vaccinated with live attenuated vaccine was significantly high at 1-5 week post vaccination, and also that of sheep vaccinated with inactivated vaccine was significantly at 2-5 week post vaccination, with less value than in animals vaccinated with attenuated vaccine. The leucocytic count of animals vaccinated with live attenuated vaccine was significantly increased at 1-5 week post vaccination and it was significantly increased at 2-5 week for animals vaccinated with inactivated vaccine. Differential leucocytic count post vaccination revealed that lymphocytic count was significantly increased after vaccination with both type of vaccines, while neutrophil count was significantly decreased after vaccination. Some kidney and liver function tests were showed non significant changes after vaccination with both type of vaccines. Total protein values had non significant changes after vaccination with both vaccines, while albumin values were showed significant decreased at 2-6 weeks post vaccination with both vaccines. Globulin value significantly increased at 1-6 week post vaccination with both vaccines. Albumin globulin ratio significantly decreased at 2-6 week post vaccination with both vaccines. SNT and ELISA tests as humural immune response showed that animals vaccinated with live attenuated RVF vaccine had protective antibodies till 24 months post vaccination, while animals vaccinated with inactivated vaccine had protective antibodies till 7 months. Maternal immunity of lambs from dams vaccinated with live attenuated RVF vaccine had protective antibodies till 4 months of age, while lamb from dam vaccinated with inactivated vaccine had antibodies till 3 months of age. Protein profile of the two viral strains showed that the most predominantly expressed polypeptide in both attenuated and virulent RVF viruses was 15 kDa.
