Predicting fertility of goat bucks by inducing acrosome reaction in sperm with calcium ionophore A23187

Abstract: Damascus and Zaraibispermatozoa were treated with 1 μmol/ml calcium ionophore A23187 to induce acrosome reactions in vitro to predict fertility of bucks in the field. Unfortunately, sperm concentration, livability and abnormality of spermatozoa are not good indicators of fertility in the field. The objective of this work is to develop an in vitro test (acrosome reaction, AR) to predict fertility in bucks spermatozoa. Semen was collected from eight goat bucks from Damascus and Zaraibi breeds, 4 bucks from each breed, once weekly for 5 collection weeks. Semen was evaluated for volume, mass motility, live and abnormal spermatozoa and sperm concentration. Calcium ionophore was used to stimulate the sperm to undergo acrosome reaction test. Results showed significant individual variation in physical semen characteristics including semen volume, sperm concentration and percentage of livability and abnormality of spermatozoa within each breed with slight breed difference in sperm concentration. Testicular measurements correlated positively with all physical semen characteristics studied, except sperm abnormality, which correlated negatively with the testicular measurements.
Percentage of live reacted spermatozoa associated with physical semen characteristics, increased by Ca ++ ionophores treatment and correlated positively with buck fertility within each breed.
It could be concluded that the fertility of goat bucks depends on the semen physical characteristics, which in turn depends on testicular measurements. Also, buck fertility could be predicted by determination of in vitro acrosome reaction in particular, when semen was treated with Ca++ ionophores.
