ُEffects of some antimetabolites chemotherapy on tumour bearing mice

Abstract: four groups of mice (grI,II,III & IV ) were subjected to the present experiment group I,II and III were uesd for adenocarcinoma implantation which were palpable 7 days post inoculation . group I, tumour- bearing mice (TB) was kept untreated Methorexate (MTX) and 5 fluorouracil (5-FU) were given to gr . II and gr III respectively , 7 days post tumour implantation (TB + MTX and TB+5-FU) group IV served as control non- tumour bearing nice (NTB) Blood and tissues samples were collected at 1and 2 weeks post trearment the plasma proteins orofile, total and differential leukocyte counts as well as oathological alterations of skin liver heart abd spleen were investigated the changes which occurred in first three groups were compared to those of the control NTB mice The changes in plasma electrophoretogram characterized by hypoproteinemia and significant decrease in mean values of albumin and globulin in TB mice However in MTX or 5-FU treated groups there were significant increase in most alpha and gamma globulin feactions the fibrinogen showed a significant decrease in both treated groups throughoutthe experiment Leukocytosis were demonstrated in - group I(TB) marked drop in leukocytes , lymphocytes neutrophils and monocytes were demonstrated in gr II (TB +MTX) and gr III (TB+5-FU ) PATHOLOGICAL INVESTIGATION of TB mice demonstared metastatic lesions in the liver ,heart and spleen The tumour cells were degenerated as a result of the chemotherapeutic agents (MTX and 5-FU ) Hapatotoxicity and depletion of lymphoid follicles in the spleen were also prominent due to the adverse effects of both drugs
