Effect of epidermal growth factor and insulin-like growth factor-ion in vitro maturation, fertilization and development of Egyptian buffalo oocytes

Abstract: This study aimed to investigate the effect of growth factors such as epidermal growth factor (EGF) or insulin-like growth factor 1 (IGF-1) alone and their combination on nuclear maturation, fertilization and development of buffalo compact oocytes cumulus cells (COCs) in vitro. Oocytes were matured in vitro for 24 h in TCM-199 medium supplemented with . 20 mmol final concentration of pyruvate and 50 μg/ml gentamicin (control medium) or supplemented with EGF (10 ng/ml), IGF-I (10 ng/ml) and EGF plus IGF-l (10 ng/ml for each). After maturation, oocytes were fertilized in vitro by spermatozoa prepared in IVFTALP medium supplemented with 20 µg ml heparin and 0.6% bovine serum albumin.
Results revealed that maturation medium supplemented with EGF plus IGF -1 combination showed the highest (P<0.05) proportion of oocytes reached metaphase-ll (M-II), cleavage rate and proportion of embryos at blastocyst stage relative to number of total matured or cleaved oocytes (72.58, 39.39, 30.06 and 76.92%, respectively) as compared to media supplemented with EGF (59.05, 30.61, 17.35 and 56.67%) or IGF-l (57.30, 25.49, 13.73 and 53.85%) and control medium (41.31, 14.81, 3.70 and 25.0%), respectively. However, supplementation of maturation media either with EGF or IGF-I showed similar effects, being higher (P<0.05) than control.
In conclusion, supplementing maturation media with EGF or IGF-l-alone and their combination led to- enhancing nuclear maturation, fertilization and subsequent development in vitro of Egyptian buffalo oocyte as compared to unsupplemented medium. The best results were obtained from medium supplemented with a combination of EGF+IGF-l as compared to either EGF or IGF-l alone.
